The effect of essential oil mixture in a mouse model of allergic asthma

Abstract
In this study, we tried to evaluate the anti-asthmatic and anti-inflammation effect of natural plant derived aromatic essential oil. Allergic asthma is a kind of immunologic disorder in respiratory epithelium, in which inflammation of airway, hypersecretion of mucus, and hypersensitivity of airway are observed. Allergic inflammation is induced by chemical mediators derived from inflammatory immune cells including T cells, B cells, mast cells and eosinophils, and chemical mediators such as cytokine or chemokine, characterized by pulmonary inflammation and increase of serum IgE level etc. To evaluate and compare the inflammatory mechanism between control group and experimental group, mouse allergic asthma model was selected. 0.025% of rosemary, marjoram, and cypress mixture (RTC-mixture) was nebulized to asthma induced mice for 8 weeks on a basis of 3 times per week and 30 min of nebulization each time. After 12 weeks from the initial experiment, the mice lungs were removed and analyzed by flow cytometer, and expressed as percentages in granulocytes/lymphocytes, CD3e+/CCR3+, CD4+/CD8+, CD69+/ CD3e+, IgE+/B220+ and CD11b+/Gr-1+cells. In the RTC-mixture treatments group, the percentages of granulocytes, CD3e+/CCR3+, CD4+/CD8+, CD69+/CD3e+, IgE+/B220+ and CD11b+/Gr-1+cells were decreased when compared with control group. In conclusion, RTC-mixture have profound effects on allergic inflammation in a murine model of asthma through suppression of granulocytes, eosinophils CCR3 expression, IgE, and CD11b cells expression.
